Core Mass Spectrometry Facility

The Core Mass Spec Facility currently has 5 Mass Spectrometry devices for your collaborative research needs. Please visit their website for more detailed information on their methods, people and instruments.

HPLC Workstations

Four HPLCs from the College of Pharmacy and Nutrition are located on Bench 1 of 2D10 and are available for public use. Ed Krol supervises and trains students on the use of this equipment. THe 4 HPLCs available include:

Waters 1 -Consists of a Waters 2695 separations module (pumps, vacuum degasser), autoinjector, column heater and 2996 UV-Vis diode array detector. This system is capable of performing both analytical and semipreparative HPLC (0.1 mL/ - 10 mL/min) and runs on Waters Empower software.

Waters 2- Consists of a Waters 2695 separations module (pumps, vacuum degasser), autoinjector, column heater and 2996 UV-Vis diode array detector. This system is capable of performing both analytical and semipreparative HPLC (0.1 mL/ - 10 mL/min) and runs on Waters Empower software.

Agilent 1- Agilent 1200 including quaternary pump, vacuum degasser, autoinjector, fluorescence detector, UV-Vis diode array detector, fraction collector. The system is capable of performing both analytical and semi-preparative HPLC (0.1 mL/min – 10 mL/min) and runs on OpenLab CDS, Chemstation Edition software.

Agilent 2- Agilent 1100 including binary pump, vacuum degasser, autoinjector, UV-Vis diode array detector. The system is capable of running analytical HPLC (0.1 mL/min – 3 mL/min) and runs on Chemstation software.

EVOS Core digital microscope

The CCHSA EVOS XL Core is a simple transmitted-light microscopy imaging system. The color camera takes images of slides that can be transported by USB stick to other locations for analysis.

LabConco Vacuum Concentrator

The CCHSA LabConco CentriVap Centrifugal Vacuum Concentrator uses a combination of centrifugal force, vacuum and heat to speed evaporation of multiple small samples. The CentriVap Cold Trap helps protect the accessory vacuum pump from the corrosive effects of vapors and fumes as they evaporate from the samples.

QX100 Droplet Digital PCR System (with droplet generator and plate sealer)

The CCHSA Biorad QX100 Droplet Digital PCR system provides an absolute measure of target DNA molecules. Applications include copy number variation, rare sequence detection, mutation detection, and gene expression analysis. The droplet generator and plate sealer needed for sample preparation are available for separate booking - please ensure you book them for the morning of the afternoon that you wish to run your ddPCR samples on the QX100. Some reagents and plates available for use on a cost recovery basis.

**The QX100 is NOT compatible with SyberGreen**

S1000 Thermocycler

The Saxena Lab's Biorad S1000 thermal cycler has gradient capabilities, with dual 48 well reaction modules that permit two separate programs to be run simultaneously.

AB 2720 Thermocycler

The Pathology & Laboratory Medicine Department's Applied Biosystems 2720 thermal cycler is a compact unit that is ideal for both basic PCR and cycle-sequencing applications using 0.2 mL reaction tubes or 96-well reaction plates.

CFX96 Real-Time PCR Detection System

The CCHSA Biorad CFX96 is a six-channel (five colors and one FRET channel) real-time PCR instrument using optical technology for singleplex or multiplex reactions for 96 well plates. The optical system, with six filtered LEDs and six filtered photodiodes, collects up to five targets of data per well. An LED filter–photodiode channel is dedicated to FRET singleplex experiments.

This instrument is located in 2D10.2.3. PLEASE NOTE:this unit cannot be used at the same time as the CFX384.

CFX384 Real-Time PCR Detection System

The CCHSA Biorad CFX384 Real-Time PCR Detection System is similar to the CFX96 system (please see above), with a block for 384 well plates. The optical system, with six filtered LEDs and six filtered photodiodes, collects up to five targets of data per well. An LED filter–photodiode channel is dedicated to FRET singleplex experiments.

This instrument is located in 2D10.2.3. PLEASE NOTE: this unit cannot be used at the same time as the CFX96.

Synergy HT Fluorescent Plate Reader

The CCHSA Bio-tek Synerg HT Multi-Detection Microplate Reader can perform fluorescence, TRF, absorbance and luminescence assays for top and bottom-read assays. It is used together with Gene 5 software for experimental design and ease of data analysis. The unit fits standard 96-well plates, and has filter wheels containing a number of filters.

Biotek EL808 Microplate Reader

There are two Biotek EL808 microplate absorbance readers in 2D10 (Department of Pathology and Laboratory Medicine and CCHSA). The EL808 is paired with Gene 5 controlling and data analysis software. The EL808 fits standard 96-well plates, and has a filter wheel containing 4 filters.

These instruments are located in 2D10 and B250.1.

Mettler MX5 Microbalance

The CCHSA Mettler MX5 analytical microbalance has a weight capacty of 5.1g with 0.001 mg readability.

The CCHSA Mettler XP26 analytical microbalance has a weight capacity of 22 g with 0.001 mg readability.

Biospec Mini-Beadbeater-24

The Mini-Beadbeater-24 disrupts microbial or tissue samples by agitating up to 24 2mL screw-cap microvials containing small glass, ceramic or steel beads and disruption buffer between 2000-3450 strokes/min. Samples are homogenized in 3 minutes in 0.1 -1 ml of medium. The non-foaming, aerosol-free method preserves enzymes and organelles and can recover high yields of DNA or RNA with appropriate solutions. Users are responsible for purchasing their own beads, solutions and sample vials.

Biorad GelDoc UV Imaging system

The Saxena Lab's Biorad GelDoc Digital Imaging System can capture UV and visible images for printing or analysis using Biorad ImageLab Software. A USB is needed to transport files since this unit not connexted to the internet. The unit is used to image both ethidium bromide and other stained gels.

This instrument is located in 2D10.2.1 (the gel/ ethidium bromide room).
